Born in about 1665 in either Cambridge or Billerica, the second Samuel Manning was raised at Billerica during the years preceding the construction of the Manning Manse. In about 1695 (he would have been about 30 years old) he moved back to Cambridge, to the old William Manning home at the corner of South and Dunster Streets, just above the banks of the River Charles, which he purchased from his father. The property included a warehouse, a boathouse, and a wharf. He became known as a "waterman", probably earning his livelihood as a merchant, dealing in goods arriving and leaving by boat.
Like his father and grandfather, he was frequently elected to office in his town. He was at various times the Sealer of Weights and Measures, a highway surveyor, Constable, Clerk, and selectman. He was the keeper of the town store of ammunition for some period, which is a further indication of the trust the citizens of Cambridge placed in him. He never returned to Billerica, and the Manse there passed to his half brother, William, upon the death of their father in 1711.
In about 1720 Samuel purchased a farm in Windham, Connecticut, and moved there. The area in which he lived was later renamed Scotland, and it is there that he died, on February 23, 1755, at about 90 years of age. He was married to Deborah Spalding at Chelmsford, Mass. on September 12, 1667. Deborah died at Windham on August 8, 1727, after bearing nine children. Samuel remarried on June 10, 1731, to Sarah Gale. This second marriage produced no children. Sarah died at Windham on October 11, 1746.
Samuel and Deborah's first three children were born at Billerica, and the rest at Cambridge. His firstborn son, again named Samuel, continues the line toward the present.
